The doctor should do a physical examination and order some imaging tests, like an ultrasound or MRI of the spine. Doing this will provide them with the information necessary to determine whether the dimple is benign and whether it is causing any other problems, like spina bifida occulta or spinal cord tethering. If the tests do not show any signs of underlying problems, the doctor will probably just monitor the markings. If anything changes, the doctor may need to run additional tests and re-examine the area.
